Mesenchymal stem cells have been shown to improve collagen productionsand regenerate the skin, for example during wound healing. These processes are mediated by vesicles known as exosomes which are produced and secreted by these stem cells. A novel active ingredient based on goji plant stem cells was shown to improve the stemness of aged mesenchymal stem cells as well as increase exosome signalling by mesenchymal stem cells, which in turn improves extracellular matrix production in fibroblasts. dehydroepiandrosterone (DHEA) play important roles in the skin. The fast that they are aise synthesised locally in the skin makes them interesting targets for cosmetic ingredients. An extract of monk's pepper (Vitex agnus-castus) was found to perform like beta-endorphin and to stimulate the synthesis of DHEA. In a clinical study, the extract showed significant effects on skin elasticity and density.Note de contenu : - Fig. 1. EnzymesUne enzyme est une protéine dotée de propriétés catalytiques. Pratiquement toutes les biomolécules capables de catalyser des réactions chimiques dans les cellules sont des enzymes ; certaines biomolécules catalytiques sont cependant constituées d'ARN et sont donc distinctes des enzymes : ce sont les ribozymes.
Une enzyme agit en abaissant l'énergie d'activation d'une réaction chimique, ce qui accroît la vitesse de réaction. L'enzyme n'est pas modifiée au cours de la réaction. Les molécules initiales sont les substrats de l'enzyme, et les molécules formées à partir de ces substrats sont les produits de la réaction. Presque tous les processus métaboliques de la cellule ont besoin d'enzymes pour se dérouler à une vitesse suffisante pour maintenir la vie. Les enzymes catalysent plus de 5 000 réactions chimiques différentes2. L'ensemble des enzymes d'une cellule détermine les voies métaboliques qui peuvent avoir lieu dans cette cellule. L'étude des enzymes est appelée enzymologie.
Les enzymes permettent à des réactions de se produire des millions de fois plus vite qu'en leur absence. Un exemple extrême est l'orotidine-5'-phosphate décarboxylase, qui catalyse en quelques millisecondes une réaction qui prendrait, en son absence, plusieurs millions d'années3,4. Comme tous les catalyseurs, les enzymes ne sont pas modifiées au cours des réactions qu'elles catalysent, et ne modifient pas l'équilibre chimique entre substrats et produits. Les enzymes diffèrent en revanche de la plupart des autres types de catalyseurs par leur très grande spécificité. Cette spécificité découle de leur structure tridimensionnelle. De plus, l'activité d'une enzyme est modulée par diverses autres molécules : un inhibiteur enzymatique est une molécule qui ralentit l'activité d'une enzyme, tandis qu'un activateur de cette enzyme l'accélère ; de nombreux médicaments et poisons sont des inhibiteurs enzymatiques. Par ailleurs, l'activité d'une enzyme décroît rapidement en dehors de sa température et de son pH optimums.

PeptidesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Skin ageing is induced by an excess of reactive chemical species caused by ultraviolet exposure, by pollutants or as adverse effects of normal cellular metabolism. Protection of the skin is mediated by the transcription factor Nrf2 which activates the expression of antioxidant and detoxification enzymes. Nrf2 is normally repressed by binding to Keap1 in the cytoplasm. In response to chemical stress, Nrf2 is released from Keap1 to migrate into the nucleous for activation of gene expression. The peptide acetyl-DEETGEF which comprises the binding sequence of Nrf2 to Keap1, can be used to disrupt the complex and this way to stimulate Nrf2 activity to enhance protection against reactive chemical species. For better skin uptake, the peptide was incorporated into solid lipid nanoparticles. A cream containing the peptide ingredient applied to skin explants was indeed found to up-regulate the expression of Nrf2-regulated genes. In skin explants exposed to UV, the peptide strongly inhibited the formation of sunburn cells and depletion of Langerhans cells. In a clinical study, the cream with a peptide ingredient showed a significant, placebo-controlled protection against UVA-induced DNA damage. Peau -- Soins et hygièneIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Today’s society is characterized by demanding jobs, long working hours and an increasingly hectic lifestyle, which frequently results in a lack of sleep. This is often markedly reflected of the skin by a tired appearance of the face. Recent research showed that also on the molecular level, lack of sleep causes stress and leads to premature aging. A major stress mechanism, the unfolded protein response (UPR) of the endoplasmic reticulum (ER), gets compromised by sleep deprivation, leading to the accumulation of misfolded proteins which damage the cell. To target this novel cellular aging mechanism, an extract of the psychrotolerant Swiss glacier bacterium Iodobacter ssp. was developed and analyzed for its efficacy to reduce ER stress and visible signs of tiredness. Treatment of aged fibroblasts with the extract led to an increase in the expression of ER chaperones which mediate the UPR and energized cells through an increase of ATP levels in a cellular model of sleep deprivation. Placebo-controlled randomized clinical studies conducted with sleep-deprived and overworked volunteers demonstrated that treatment with the Iodobacter-derived active ingredient IceAwakeâ„¢ improved several skin parameters associated with skin aging, leading to a vitalized and rejuvenated appearance.
