Titre : Immobilization of degradative enzyme onto collagen hydrolysate films Type de document : texte imprimé Auteurs : C. Shanthi, Auteur ; B. Stennett, Auteur ; Dennis C. Shelly, Auteur Année de publication : 2003 Article en page(s) : p. 6-12 Note générale : Bibliogr. Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Chemical or biological degradation of chrome shavings results in collagen hydrolysate and chrome cake. To add value to the recovered collagen hydrolysate, we evaluated films, cast from this material, for immobilizing degradative enzymes. Organophosphorus hydrolase (OPH) has been shown to be effective in degrading a range of organophosphate esters. Organophosphate compounds, which are among the most toxic substances known, are used as pesticides and nerve gases. A crude extract of OPH from Flavobacterium sp. was immobilized onto collagen hydrolysate films. The collagen films were characterized by differential scanning calorimetry and water swelling behavior. The properties of the collagen hydrolysate films were compared with the films cast from commercially available gelatin. The effect of glutaraldehyde crosslinking the films on enzyme immobilization was studied. Spectrométrie infrarougeIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : The ability of such alkalis as calcium, barium, sodium and potassium hydroxides and sodium metasilicates to immunize wool was investigated. Sodium silicate was confirmed as an effective immunization agent whose efficiency increases with inceased tratment duration and treatment solution concentration. Sodium silicate lacks a divalent cation and acts at pH13. This allows the assumption that the immunization mechanism is other than that of calcium or barium hydroxide. Differential scanning calorimetry and infrared spectroscopy analysis results show the differences in hair structure after treatment by various alkalis. Research work is in progress to clarify the mechanism of the hair immunization by sodium silicateand to develop a unhairing processwith hair immunizationwhich is suitable for leather processing En ligne : https://drive.google.com/file/d/13id5nFtYv_VWJqo-W-x1DuFlB4GZ2Xpl/view?usp=drive [...] Format de la ressource électronique : Permalink : https://e-campus.itech.fr/pmb/opac_css/index.php?lvl=notice_display&id=24648

Silicate de sodiumIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : A major contributor to the pollution from the tanning industry is conventional unhairing using sodium sulphide and lime. In this study, an attempt has been made to develop a lime-free unhairing process using sodium silicate as a hair immunization agent. The parameters of the unhairing process have also been optimized evaluating qualitative indexes of the hide during processes and the leather obtained as well. It was established that hair removal quality and chemical and strength properties of the experimental leathers are comparable to those control leathers. A significant reduction in chemical oxygen demand (COD), biochemical oxygen demand and total Kjeldahl nitrogen were obtained in comparison with the comparative data for the conventional unhairing with hair burning. Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : An indirect sandwich ELISA procedure has been developed for estimation of the decorin core-protein content of preparations of bovine decorin. A mouse anti-bovine decorin antibody is used as the capture antibody, and a rabbit anti-bovine decorin antibody as the detecting antibody. Goat anti-rabbit immunoglobulin G (IgG) labeled with horseradish peroxidase is used to react with the detecting antibody. The peroxidase substrate orthophenylenediamine (OPD), when exposed to the (bound) enzyme in the presence of peroxide, yields a soluble product with an absorbance maximum (in acidic medium) at 492 nm. The absorbance of the solution at this wavelength thus reflects the amount of decorin core-protein in the decorin preparation that is recognized by both the capture antibody and the detecting antibody.
Note de contenu : - PRINCIPE DE FONCTIONNEMENT DES SYSTEMES TESTES : Palisson humide - Sèche sous vide à basse température
