The aim of the present study was to investigate the anti-dandruff and sebum regulating potential of an extract of Epilobium angustifolium. In vitro the extract (0.1 %) inhibits testosterone-induced 5-alpha reductase activity in fibroblast by 15.7 °/0 and reduces lipid synthesis in sebocytes by 43%. In vitro, E. angustifolium extract also dose-dependently increases involucrin expression in keratinocytes. Treatment of skin explants with E. angustifolium extract (1.5 %) protected the epidermis from Malassezia-induced formation of pyknotic nuclei. As shown ex vivo, E. angustifolium extract acts through innovative mechanisms involving modulation of the expression of defensins (hBD2 and hBD3) and TLR2 acting at the interface of innate and adaptive immune responses. The anti-dandruff and sebum regulating efficacy of E. angustifolium extract were confirmed by two clinical studies whose results will be presented in a subsequent issue of SOFW. In conclusion, E. angustifolium extract offers a new innovative approach to dandruff reduction through immunomodulation of the skin response to Malassezia invasion. |
