This makes the investigation of the fibrillar collagen type characteristics interesting for skin research. In this study collagen types were characterized using 5-dimensional multiphoton laser scanning microscopy (5D-IVT) that can be applied in vivo. Second harmonic generation (SHG) signals and fluorescence lifetimes of the collagen autofluorescence were analysed. Collagen type I generates a higher SHG intensity and a longer fluorescence lifetime compared to collagen type III. Thus, the SHG intensity decrease found in photodamaged skin might be explained by the increase in collagen type III. Calculating the in vivo relevant increase of collagen type III gives a negligible difference in fluorescence lifetime not qualifying this method for the determination of collagen type changes in dermal photodamage in vivo in human skin. However, for pathologies that exhibit higher differences in collagen types 5D-IVT analysis might be a suitable method. Photovieillissement (dermatologie)Index. décimale : 668.5 Parfums et cosmétiques Résumé : Specific visualization and assessment of human collagen fibers were demonstrated using a developed second-harmonic-generation (SHG) microscope equipped with a Cr:forsterite laser (1250 nm). Initially, we examined the specific sensitivity of SHG imaging for collagen fibers using excised human skin specimens. Age-related dermal alterations in female subjects were then evaluated by means of SHG microscopy. The developed SHG microscope enabled us to observe the collagen fibers in the skin sections ; their SHG images correlated well with the Elastica-van-Gieson-stained sections of dermal fibers. In vivo imaging of human skin with a wide field of view as also achieved. En face SHG images of facial skin of female subjects of different ages showed the structural alterations in the collagen fiber network. Additional image analysis of SHG images based on two-dimensional Fourier transformation also reflected age-related alterations. In conclusion, this proposed methodology can be a powerful tool in fields such as anti-aging dermatology and validation of the efficacy of cosmetic products.
