Stress -- Effets physiologiquesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The continuous research for new, high-performing ingredients is one of the most difficult hurdles facing cosmetic companies. Sometimes, a more in-depth investigation on the structures composing the apparently homogeneous phases of cosmetic preparations is required. This could allow a precise understanding of some hidden molecular behavior. This paper describes an innovative approach for the identification of skin-effective raw materials. It is based on the combined application of in silico analysis of molecular dynamics followed by a new in vitro model for studying the skin recovety from stress. This new pathway was adopted to better understand the molecular structure of the aqueous phase of cosmetics containing osmotic protectants and their mechanism of interaction with the protein moieties when some type of stress conditions is applied.
Molecular dynamics is a method using the computer simulation of the physical movements of atoms and molecules in a specific aqueous environnent. The in vitro stressed skin cell test, which was successively applied to the aqueous solutions, revealed through specific biochemical markers the different effects of some water binding molecules on skin protection and recovery.
This new approach was eventually con-firmed by in vivo study models. The testing protocol we applied was the one which is generally used for photo-aging studies: the microrelief and the elasticity of skin submitted to UV and surfactant stress were determined alter prolonged use of the product by volunteers and a microstructure evaluation of the dermoepidermal junction was carried out.
The in vivo results obtained for some bio-physical parameters of the skin of aged volunteers were in agreement both with the theoretical (in silico) and biological (in vitro) findings. Moreover, the results show that, as a consequence of their chemical structures, osmolytes exhibit different aggregation states in aqueous solution and, when properly combined, show synergistic effects in the protection of the native folded structure of proteins.Permalink : https://e-campus.itech.fr/pmb/opac_css/index.php?lvl=notice_display&id=19305

Toxicologie cellulaireIndex. décimale : 667.2 Colorants et pigments Résumé : Several benzo[a]phenoxazine dyes containing a maleimide moiety have been synthesised and characterised by proton nuclear magnetic resonance spectroscopy and mass spectrometry. The spectroscopic properties, such as absorption and emission spectra, of these dyes – 9-maleimido-5H-benzo[a]phenoxazin-5-one (7a), 6-chloro-9-maleimido-5H-benzo[a]phenoxazin-5-one (7b), and 6-bromo-9-maleimido-5H-benzo[a]phenoxazin-5-one (7c) – were examined. These compounds were evaluated as potential fluorescent and colorimetric probes for the detection of biothiols. Results show that the studied compounds exhibit colorimetric and fluorescent responses to l-cysteine at pH 7.4. In the presence of l-cysteine, the fluorescence intensity of synthesised dyes is greatly enhanced. The addition of l-cysteine to solutions of dyes 7a to 7c also results in red shifts of their absorption bands. The cytotoxic effect of tested dyes 7a to 7c against human neuroblastoma cells (SH-SY5Y) was determined by MTT assay. The fluorescent response of these compounds to thiols SH-SY5Y was examined. The results show that the tested compounds exhibit fluorescent response in this cell line, and the benzo[a]phenoxazine dyes substituted in the 6-position with a halogen (7b and 7c) are less toxic against cells under experimental conditions than their unsubstituted analogue (7a). Tests de sécuritéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Contexte : Une étude systématique et détaillée a été conçue et réalisée, en tenant compte de certains des avantages proposés, comme une plus grande efficacité, la transparence, la texture unique, la protection de l'ingrédient actif et une plus grande acceptabilité par les consommateurs, etc., des produits cosmétiques contenant des oxydes métalliques nanométriques.
Methodes : La présente étude décrit également un procédé in vitro pour déterminer le facteur de protection solaire des échantillons de crème écran solaire expérimentales contenant de l'oxyde de zinc et de dioxyde de titane avec une gamme variée de taille des particules. Enfin, une étude comparative a été réalisée entre les particules d'oxydes métalliques classiques, ainsi que des nanoparticules.
Resultats : Tous les produits formulés ont été thermiquement stables, ayant un pH allant de 7.9 à 8.2. En outre, les teneurs en acide gras et de résidus ont été trouvés être analogues aux valeurs standard dans chaque cosmétique. La formulation contenant des nanoparticules d'oxyde de zinc a été trouvée plus facile à étaler par rapport à des produits cosmétiques contenant des particules d'oxyde de zinc classiques, respectivement. De même, la crème E/H contenant les nanoparticules de dioxyde de titane a été trouvée avoir plus spreadabilty par rapport à des produits cosmétiques pour la peau contenant des particules de dioxyde de titane conventionnels. Toutes les formulations se sont révélées avoir une distribution uniforme des particules. Les crèmes de protection solaire contenant des nanoparticules d'oxyde de zinc et les nanoparticules de dioxyde de titane se sont révélées avoir des facteurs de protection solaire in-vitro plus élevés (SPF de 3.65 pour les nanoparticules de ZnO et de 4.93 pour les nanoparticules de TiO2) par rapport à celle de la crème de protection solaire contenant des particules d'oxyde de zinc classiques (SPF = 2.90) et le dioxyde de titane classique (SPF = 1.29), indiquant clairement l'effet de la réduction de la taille des particules, du micro au nano, sur le facteur de protection solaire.
Conclusion : Une bonne texture, un meilleur étalement et un SPF in vitro amélioré prouvent le rôle avantageux de nano- particules dans les produits cosmétiques.Note de contenu : - Characterization of zinc oxide and titanium dioxide nanoparticles

Les propriétes olfactives de tous les esters obtenus ont été determinées; les esters aromatiques sont inodores, alors que les esters aliphatiques ont montré des propriétes intéressantes. Certains d'entre eux délivrent des notes fruitées, vertes, boisées ou encore de pin ou de violette, alors que d'autres pourraient être des arômes agroalimentaires. Des tests de toxicitéin-vitro ont été menes sur l'ester cyclopropylique, qui est le plus prometteur de ces composants. Ce sont les kératinocytes humains NCTC 2544 et les fibroplastes de souris L-929 qui ont été utilisés pour cette étude; les résultats obtenus montrent que l'ester cinéolique étudié est beaucoup moins cytotoxique que le dodécylsulphate de sodium, utilisé comme témoin.
