CaroténoidesLes caroténoïdes sont des pigments plutôt orange et jaune répandus chez de très nombreux organismes vivants. Liposolubles, ils sont en général facilement assimilables par les organismes. Ils appartiennent à la famille chimiques des terpénoïdes, formés à partir de la polymérisation d'unités isoprènes à structure aliphatique ou alicyclique. Il est généralement admis qu'ils suivent des voies métaboliques similaires à celles des lipides. Ils sont synthétisés par toutes les algues, toutes les plantes vertes et par de nombreux champignons et bactéries (dont les cyanobactéries).

Stress oxydatifIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The availability of innovative noninvasive methods for the determination of free radicals and antioxidants in human skin allows new insights into the interaction process of free radicals and antioxidans. Summarizing the results of different studies shows that high levels of antioxidants represent an efficient prevention strategy against skin aging. The level of antioxidants in the human skin is strongly influenced by the lifestyle and stress conditions of the volunteers. The investigations were carried out using a noninvasive optical Raman spectrometer and a skin scanner for noninvasive remission measurements. In addition, electron paramagnetic resonance measurements were used to analyze both the radical formation in the skin after exposure to external stress factors and the antioxidative protection status. In a one-year study it could be demonstrated that the kinetics of the antioxidants in the human skin reflect the lifestyle of the volunteers. The antioxidatide status is strongly influenced byt the nutritional behavior and stress conditions. Analysis of skin aging ant the antioxidant status of the volunteers, all of whom were around 50 years of age and had not changed their lifestyle and nutritional behavior over the last decades, dmonstrated that a high level of antioxidants is the best protection strategy against skin aging. How these antioxidants can penetrate the skin best is described in the present article. For this purpose, the systemic and topical application of antioxidants is compared.
Taking into consideration the interaction between antioxidants and free radicals in human skin, the question arises whether sunscreens should provide protection against radiation not only in the UV but also in the visible and infrared solar spectra. "Sun protection" initiated a multicenter study in accordance with the in vivo test protocol of the International Sun Protection Factor (SPF) Test Method. Three different amounts (0.5, 1.0 and 2.0 mg/cm2) of three commercial sunscreens were investigated in three test centers. The main result obtained was a linear dependence of the SPF on the quantity applied. The recommendation to use an application quantity of 2.0 mg/cm2 is clearly driven by the aim to produce reliable and reproducible in vivo resluts. In the subsequent in vitro multicenter study, possible methodologies and strategies for improved accuracy of the determination of the SPF on sandblasted PMMA plates were compared. Influencing factors such as the amount of product applied, pretreatment of the samples, equilibration time and temperature, and the quality of the spectrophotometer were taken into account. An overall decrease in the standard deviations within single laboratories could be achieved for in vitro SPF testing but no improvement in the span of interlaboratory differences was obtained.
The complete findings obtained from this multicenter study could perhaps serve as a solid basis for increasing the reliability of a future in vitro SPF method.Note de contenu : - In vivo multicenter study

Vieillissement cutanéIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Vitreoscilla filiformis (Vf) est un microorganisme filamenteux qui est endémique de certaines eaux de source. Cette bactérie a été identifiée comme un contributeur probable des effets bénéfiques de certaines eaux thermales sur la peau. Nous avons évalué une fraction de la biomasse de Vf produite à grande échelle par un procédé biotechnologique pour sa capacité à moduler l'expression des ARN messagers des cellules cutanées in vitro. A l'aide d'un pré-criblage sur puces à ADN incluant une sélection de 1176 gènes totalement identifiés et représentatifs de la physiologie cutanée et de son homéostasie puis du système de puces U133 plus 2.0 pour le génome complet d'Affymetrix nous avons identifié la Manganèse Superoxyde Dismutase mitochondriale (MnSOD/SOD-2) comme une cible très préférentiellement induite dans les fibroblastes dermiques et les biratinocytes épidermiques humains normaux. Cette induction transcriptionnelle a été confirmée et quantifiée au moyen de la RT/PCR quantitative. Une étude de cinétique d'expression a montré un pic d'expression de MnSOD après 20 heures de stimulation. Au moyen de l'analyse par immunofluorescence (FACS), nous avons constaté, conformément aux résultats d'expression obtenus, une induction de synthèse protéique dans les fibroblastes dermiques humains normaux (×1.6; P < 0.01) et les kératinocytes humains normaux (×1.4; P < 0.01). Comme MnSOD est l'enzyme antiradicalaire inductible majeure des cellules cutanées, nous suggérons que cette fraction biotechnologique sélectionnée de Vf peut induire les cellules cutanées à produire leur propres systèmes de défense endogène in vivo. Comme attendu de ces observations, le traitement de la peau in vivo chez l'homme avec un extrait de Vf a montré une protection significative en termes d'apparition des cellules coup de soleil (sunburn cells) dans l’épiderme, une signature possible des effets délétères du soleil sur la fonction antioxydante mitrochondriale MnSOD cutanée. L'ensemble de ces observations suggère l'intérêt d'associer cet extrait bioactif de Vitreoscilla filiformis avec des filtres solaires adaptés afin de potentialiser la protection cutanée par des voies complémentaires.
