Peroxyde d'hydrogèneLe peroxyde d'hydrogène (H2O2), communément appelé eau oxygénée ou encore perhydrol (appellation industrielle), est un composé chimique liquide et visqueux, aux puissantes propriétés oxydantes (il est aussi réducteur). C'est donc un agent blanchissant efficace qui sert de désinfectant et (à haute concentration) d'oxydant ou monergol dans les fusées spatiales.

In this study, both H2O2 (450μM) and UVC (40J/m2) irradiation-induced cell death were detected by MTT assay, and nicotinamide showed the dose dependent inhibition on H2O2 and UVC induced cell death. Morphological study by microscopic observation showed that H2O2 or UVC induced cellular morphological changes including appearence of rounded cells or swelling cells, characteristic og apoptosis. In the presence of nicotinamide, H2O2 and UVC induced morphological changes were attenuated significantly. Prevention of cells from H2O2 and UVC induced cell death was asociated with decrease of apoptotic bodies by flow cytometry analysis and attenuation of the appearance of DNA ladders by DNA electrophoresis. Induction of p53 protein was detected in H2O2-treated NIH3T3 cells and nicotinamide suppressed its increase. However, bcl-2, bax and α-tubulin proteins were remained unchanged. ROS have been demonstrated to be involved in the H2O2 and UVC induced apoptosis. In this study, H2O2 or UVC irradiation increase intracellular peroxide levels detected by DCHF-DA assay and fluorescent microscopic observation, and nicotinamide blocked the peroxide level induced by H2O2 or UVC irradiation. Decrease of H2O2 or UVC-induced 8-OH dG in DNA was also detected in nicotinamide trated cells. The results of present study firstly demonstrated that nicotinamide prevented cells from H2O2 and UVC irradiation induced apoptosis through suppression ROS production, p53 elevation and 8-OH dG formation.
