Though emollients are commonly used, little is known about their effects on the biomechanical barrier properties of human stratum corneum (SC), which play a critical role in consumer perception of formulation efficacy. Accordingly, our objective was to provide new insights with a study involving fourteen cosmetic emollient molecules with widely varying structures, molecular weights, SC diffusivities, topological polar surface areas (TPSAs), viscosities and chemical functionalities.
- Methods : Mechanical stress in the SC was measured in vitro using a substrate curvature measurement technique. Stress development due to SC drying was measured before and after topical treatment with cosmetic emollients. Emollient diffusivity and alterations to lipid content in SC after treatment were measured via ATR-FTIR spectroscopy. The maximum penetration volume of emollient in SC was characterized to elucidate mechanisms underlying emollient effects on stress.
- Results : The application of all cosmetic emollients caused a reduction in SC mechanical stress under dehydrating conditions, and a linear correlation was discovered between emollient penetration volume and the degree of stress reduction. These molecules also induced increases in stress equilibration rate, signalling changes to SC transport kinetics. Stress equilibration rate increases linearly correlated with decreasing intensity of the νCH2 band, indicating a previously unknown interaction between cosmetic emollients and SC lipids. Stress and penetration volume results were rationalized in terms of a multi-parameter model including emollient molecular weight, diffusivity, TPSA and viscosity.
- Conclusion : We provide a new rational basis for understanding the effects of cosmetic emollient choice on biomechanical properties affecting SC barrier function and consumer perception. We demonstrate for the first time that emollients very likely reduce SC mechanical stress through their ability to take up volume when penetrating the SC, and how molecular weight, SC diffusivity, TPSA and viscosity are predictive of this ability. As cosmetic formulations continue to evolve to meet the needs of customers, emollient molecules can be selected that not only contribute to formulation texture and/or spreadability but that also leverage this novel connection between emollient penetration and SC biomechanics.Note de contenu : - Stratum corneum preparation

Pollution -- Effets physiologiquesIndex. décimale : 668.5 Parfums et cosmétiques Résumé : The World Health Organisation (WHO) estimates that more than 80% of people living in urban areas are exposed to air quality levels that exceed defined health limits. In addition to solar ultraviolet radiation (UVR), noxious gases, particulate matter, and ozone, which are all found in or generated by combustion smoke, are key air pollutants that have a negative impact on the skin. An indoor combustion smoke with frequent contact on humans is cigarette smoke. Smoking is associated with early skin ageing (wrinkles, skin dryness, skin discoloration, etc.), and the combination of smoking and solar UVR has been reported to further exacerbate skin ageing processes. Using cigarette smoke as a pollutant model, this paper describes a new validated clinical method to evaluate the "protective" effects of specific cosmetic products against gaseous pollution exposure. The skin of human volunteers is treated with the product under test, exposed to cigarette smoke and then peroxidation of human sebum or skin barrier lipids is assessed using LC-MS. Key advantages of this method are the small number of volunteers required, and the reproducibility and sensitivity of the method to detect protective and antioxidative properties of cosmetic active ingredients and cosmetic formulations. Pollutant cigarette smoke is a suitable substance containing all key pollution components that can be used in human volunteers without ethical issues as compared to other pollution sources. Furthermore, this method enables us to induce, under standardised clinical laboratory conditions, pollution stress on "living skin" in healthy subjects. StatistiqueIndex. décimale : 668.5 Parfums et cosmétiques Résumé : Walnut green husk is one of the main waste products from walnut and could be used as a source of natural dyeing compounds such as juglone. The present study was conducted to evaluate the effective use of walnut green husk extract as a natural hair dye. Dyeing properties, fastness and antimicrobial behaviours of dyed hair and also a skin irritation test for natural hair dye on rat skin were examined. When the extract was mixed with ascorbic acid as a developer, ferrous sulphate as a mordant, and Aloe vera extract used as a secondary mordant and also a cosmetic ingredient, the reaction resulted in a dark-brown colour on hair samples. The dyed hair exhibited appropriate colour strength having excellent morphology for a hair surface coated with dye molecules. In addition, the dyed hair possessed good resistance to washing and daylight fastness, without any irritant properties as shown in a rat model, although high concentrations of iron-based mordant may be problematic for long-term usage. This paper also suggests the use of natural mordants such as lactic and oxalic acids to avoid any probable risks. Walnut green husk extract was an appropriate natural hair dyeing agent in practice and showed maximum antimicrobial activity compared with semi-synthetic and commercial hair dyes. The results demonstrated that walnut green husk can be used as an economical, valuable, eco-friendly and safe source of dyeing and antimicrobial agents for cosmetic products.
Note de contenu : - MATERIALS AND METHODS : Biochemical analysis
