Glucose oxydaseLa glucose oxydase (GOx, GOD) est une enzyme oxydo-réductase (EC 1.1.3.4) qui catalyse l'oxydation du glucose en peroxyde d'hydrogène et en D-glucono-δ-lactone. Dans les cellules, elle participe à cliver les sucres (oses), notamment le saccharose (Glc-Fru) en métabolites.
La GOx est largement utilisée pour déterminer la concentration en glucose libre dans les fluides corporels (diagnostic), et dans les aliments (industrie). Elle a de nombreuses applications en biotechnologies, typiquement les tests enzymatiques de biochimie.

RevêtementsIndex. décimale : 667.9 Revêtements et enduits Résumé : Trace oxygen in packaged foods, beverages, and pharmaceuticals can promote a range of oxidative degradation reactions and support microbial growth, ultimately impacting product quality and shelf-life. Oxygen scavenging active packaging systems have therefore been explored to control headspace oxygen content. Herein, we report on a hydrophobic ion pairing method to render glucose oxidase hydrophobic, and thus soluble in organic solvents. Hydrophobic modified glucose oxidase was blended with ethylene vinyl acetate and cast on the interior of glass vials to demonstrate potential as a commercially translatable coating method for enzyme immobilization. The resulting oxygen scavenging polymer coatings were topographically uniform and presented 0.553 μg/cm2 enzyme at the coating interface. The coatings effectively reduced headspace oxygen by 2% in a closed-vial system filled 50 vol% with citrate buffer, pH 3.5. Less than 25% protein migrated from the coating over an 8-week leaching study, with no detectable protein leached in the first 4 weeks. Hydrophobic ion pairing of glucose oxidase enabled a facile, high-throughput enzyme immobilization technique without use of complicated, time-consuming surface modification chemistries and reagents. Such oxygen scavenging polymer coatings can support controlling headspace oxygen in packaged goods, and thus retaining stability of oxygen-sensitive components such as colors, flavors, and nutrients. Formaldéhyde -- EliminationIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Samples of collagen extracted from phosphonium tanned leather shavings by three different methods (acid, alkali, magnesium oxide dissolution and alkaline protease hydrolysis treatments) were characterized with respect to their components and morphology of fibril network. The ratio of collagen gained was 90% by the process of alkali dissolution and enzyme hydrolysis, which was much higher than that of acid and alkali methods. The results from amino acid analysis showed that the components of extracted collagen were similar with that of commercial gelatin. The Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE) results indicated that the extracted substance was the mixture of polypeptide and amino acid. The polypeptide and amino acid exposed to hydrophilic functional groups including carboxyl, hydroxyl, amino group etc. These functional groups especially amino group can react with formaldehyde which leads to scavenge formaldehyde. The morphological change of leather shavings treated by magnesium oxide and alkaline protease was clearly presented with scanning electron microscope. The percentage of formaldehyde removal and thickening rate of collagen were 51.8% and 10.4%, respectively. Therefore, the collagen from the leather shavings and used as the formaldehyde scavenger has a good potential in achieving the innocent treatment and reuses of the solid leather wastes. RhodamineIndex. décimale : 535.6 Couleur Résumé : We designed a rhodamine B-based colorimetric chemosensor BHSO ((Z)-3′,6′-bis(diethylamino)-2-(2-(((8-hydroxy-2,3,6,7-tetrahydro-1H,5H-pyrido[3,2,1-ij]quinolin-9-yl)methylene)amino)ethyl)spiro[isoindoline-1,9′-xanthen]-3-one) for detecting Cu2+. In the presence of Cu2+, BHSO caused a colour variation from colourless to bright orange. The limit of detection for Cu2+ towards BHSO was 0.73 μM. The binding of BHSO to Cu2+ was analysed as a 1:1 ratio through a Job plot and electrospray ionisation mass spectrometry. BHSO can detect readily Cu2+ with a test strip by colorimetric variation. The detecting process of Cu2+ by BHSO was represented by ultraviolet-visible titration, electrospray ionisation mass spectrometry, proton nuclear magnetic resonance titration, Job plot and density functional theory calculations. APTS showed a clear colour change from colourless to yellow with Cu2+. The detection limit of APTS for Cu2+ was 0.52 µmol/L. APTS could also be used as a test strip for detecting Cu2+. To understand the binding mode of APTS with Cu2+, ultraviolet-visible spectroscopy and electrospray ionisation-mass spectrometry analyses were conducted. The sensing mechanism of sensor APTS towards Cu2+ was proposed to be the combination of intramolecular charge transfer and ligand-;to-metal charge-transfer through theoretical calculations.
