The dyes were tested as fluorescent DNA intercalating probes to apply in DNA gel electrophoresis. The DNA samples stained with all dyes from the series demonstrated bright fluorescent signals. DNA fragments were successfully visualised under orange and green filters as well as under standard UV transillumination. Two of the studied dyes revealed higher sensitivity to DNA when compared with the commercial dimeric cyanine dye TOTO-1. Their sensitivity reached that of the commercial dimeric cyanine dye YOYO-1, but the emission was shifted to longer wavelengths. These qualities make the dyes suitable to apply in a wide range of medical and scientific analytical methods.

They are known to play essential roles in maintaining homeostasis and preventing infectious pathogens. In recent years, cosmetic product development has begun to focus on the relationship between skin microbiomes and skin conditions. However, the statistical methods used in many studies include the standard t-test and small-scale correlation analysis, which do not take into account the internal correlation structure in data on skin microbiomes and skin features. In this study, we aimed to understand the relationship between skin microbiomes and skin features by analysing complex microbiomes and skin data.
- Methods : We obtained data on 19 skin characteristics and skin microbiomes based on 16s ribosomal RNA (16S rRNA) gene analysis of 276 healthy Japanese women. We then performed the principal component analysis (PCA), a method that takes into account the internal correlation structure, on 234 panels of them that did not contain outliers or missing values. We confirmed the relationship between skin microbiomes and skin features with principal component regression analysis and hierarchical clustering analysis (HCA).
- Results : The principal component regression analysis showed strong relationships between skin microbiomes and sebum-related skin characteristics and skin pH. In the HCA, the female panel was classified into two major groups based on the skin microbiome. Furthermore, there were significant differences in sebum-related skin characteristics and the way skin condition changes with ageing between those groups, suggesting the possibility of measuring skin condition and age-related skin risk based on microbiome data. In addition, sebum-related characteristics differed significantly among middle-aged participants, suggesting a strong relationship between skin microbiomes and sebum-related characteristics.
- Conclusion : Analysis of skin condition and skin microbiome in Japanese women, taking into account the correlation between variables, showed that skin microbiome was significantly related to the number of pores and the amount of sebum. Furthermore, it was suggested that the skin condition and the way the skin condition changes with ageing may differ depending on the type of skin microbiome. The finding of a relationship between skin condition and skin microbiome suggests the possibility of proposing a new beauty method focusing on the skin microbiome in the future.Note de contenu : - SUBJECTS AND METHODS : Study population - Acquisition of data on skin features - Acquisition of skin microbiome data and calculation - Computational analysis - Selection of microbiome genera for analysis - Statistical analysisPrincipal component analysis - Multivariable linear regression on skin features and skin microbiomes - Hierarchical clustering analysis and Mann–Whitney's analysis on skin features and skin microbiomes

Tests biologiquesIndex. décimale : 667.3 Teinture et impression des tissus Résumé : Some test strips have high feasibility for the quick and approximate quantification of the biochemical substances in samples. Recently, test strips became more precise and accurate by involving the quantitative observation of coloration and regression modelling. Their accuracy and precision can be significantly improved by hue-shifting the colours of the original sample and those of the standards. The standard colour images included in the instructions of test strips for the detection of four urinary and two salivary biochemical substances were used. Images of glucose-loaded test strips for urinary glucose detection were also prepared. Using the red-green-blue (RGB) colour image, the author prepared 11 hue-shifted colour images by changing the hue of the original RGB colour image at intervals of 30°. From each of the 12 colour images, 10 greyscale images indicating values of the intensity of RGB, cyan, magenta, yellow, key black, L*, a* and b* were prepared. In regression modelling, the additional 110 greyscale images improved the values of the coefficient of determination and normalised mean absolute error in the quantification of protein, specific gravity, urobilinogen and glucose in urine, as well as of alcohol and nitric oxide in saliva. Further analyses showed that the hue-shifting technique yielded new greyscale images that were more significantly related to the values of the biochemical variables in question than any previous greyscale images. This improvement was realised by the most significant linear measures for the description of concentrations of biochemical substances, and synergetic effects between significant greyscale images.
