Tree-dimensional structures of a fragment of Type I collagen triple helices and microfibrils were examined to predict the formation of stabilizing interactions and binding sites for chromium and acidic crosslinkers.Interactions of arginine and lysine side chains with negatively charged side chains, which may contribute to the stability of triple helical and larger collagen assemblies, were identified in the computer models.Lysine to lysine distances were measured and used to predict suitable chain lengths for bifunctional acidic crosslinkers.Fourteen sites composed of acidic side chains separated by 6-8A suitable for crosslinking with a 7A binuclear chromium complex were identified.After conversion of amide side chains to the corresponding acid form, as would occur by liming, an additional nine sites were identified. |
