It has been established that enzymatic unhairing is an environmentally friendly alternative to the conventional process. The enzyme, an alkaline protease (clarizyme) is produced by aspergillus flavus isolate using solid substrate fermentation (SSF). Since SSF has distinct economic advantages over conventional methods, it is hoped that this alkaline protease will be available at a commercially attractive price. Evolution of this enzyme as a depilating agent for goat leather establishes that the enzymatically dehaired skins have better strength compared to the controls. This paper highlights the process development and scaling-up strategies adopted in the enzyme production and its application. Tannage Résumé : The sludge biomass, composed largely of organic matter 60-90%(W/V), is contained within the cell wall membranes of microbial cells. It was found that the pressure pulses produced by an ultrasonic homogeniser significantlyinfluenced the degree of disintegration of the cells containing the waste. The mechanical sludge disintegration was investigated with an ultrasonic homogeniser operated at a frequency of 20 kHz with different energy intensities. The influence of sludge solids content on the disintegration of the sludge also was investigated. It was found that the influence of mechanical disintegration on particle size cannot fully describe the release of organic material into the sludge water. The release of organic material into the sludge water can best be estimated by the chemical oxygen demand (COD) and by protein release. The degree of disintegration, as indicated by COD and protein release, increased significantly when the energy intensity applied with the ultrasonic homogenizer was increased. DésorptionIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Preservation of animal skins by conventional salt or brine curing methods leads to environmental pollution. Controlled drying up to acceptable moisture levels could be an effective, cleaner and environmentally friendly alternative for skin preservation. The selection of optimum storage and drying conditions requires the knowledge of moisture desorption isotherms. Hence this work focuses on the experimental determination of desorption isotherms for south Indian sheep and goat skins at three different temperatures 30, 40 and 45°C. The goodness of fit of four sorption models to experimental results was determined. Of the models tested, the modified Oswin gave the best fit followed by modified Halsey. Modified Henderson and modified Chung Pfost models showed relatively larger deviations from the experimental values. The deviation was larger in sheep skins compared to goat skins. The net isosteric heat of sorption was determined for both sheep and goat skins using the Clausius - Clapeyron equation and is expressed mathematically as an empirical polynomial function of equilibrium moisture content. The net isosteric heat of sorption for sheep skin was higher than the goat skin.
