One can not only observe the various entities and study their morphology but also follow the reactions that a substance undergoes when subjected to several treatments.
The advantages of modern electron microscopy include, apart from the require ment of small quantities of sample, stereoscopic view, possibility of heating or cooling the sample, and observation of the specimen under moist conditions.
The results obtained from electron microscopical analyses can broadly be discussed under the following headings :
(a) Raw material-Collagen
(b) Tanning materials
(c) Effect of pre-tanning and tanning operations on collagen
(d) Other modifications such as irradiation

Wet-blue (tannage)Peau tannée au chrome (le chrome donne une couleur bleue)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Inductively Coupled Plasma Optical Emission Spectrophotometer (ICP-OES) was used to accurately determine the Chromic Oxide content in 90 total Wet Blue, Crust, and Finished Leather samples. Accuracy was verified on each sample by testing retained composite material using ASTM D2807 Perchloric Digestion / Sodium Thiosulfate Titration Method. The ICP-OES Method correlated to ASTM D2807 with less than 2% deviation from the averaged mean value and less than 3% deviation from the averaged standard deviation value. The Chromic Oxide mean values for ASTM D2807 and ICP-OES were 3.11 ± 0.70% and 3.17 ± 0.68% respectively. The new method maintains a highly consistent statistical capability with Cpk at 2.15 vs. 2.17. Oxidation of chromium in leather samples is not required for chromic oxide detection. Perchloric acid more than doubles the required consumables cost, test time and hazard required to validate chromic oxide levels in leather materials. The ICP-OES method eliminates the crystal formation explosion hazard and chlorinated waste environmental hazards associated with Perchloric acid. Replacing ASTM D2807 with the proposed method by tannery validation laboratories is cheaper, faster, safer and more environmentally sustainable than titration by our classic technique.

Wet-blue (tannage)Peau tannée au chrome (le chrome donne une couleur bleue)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : To characterize the reacting performances of proteases against chrome-tanned leather, the methods to quantitatively measure protease activities against chrome-tanned elastin and collagen fibers were established through detecting the produced amount of Desmosine (DES) or Hydroxyproline (Hypro) in unit time. Chrome-tanned elastin and collagen fiber substrates with different Cr content were prepared, and the influence of the Cr content on the enzymatic hydrolysis resistance of tanned substrates was investigated. The activities of several typical proteases against chrome-tanned elastin and collagen fibers were tested based on the methods, and the relationship between the activities and bating effectiveness on wet blue was also preliminarily investigated. The results showed that the two kinds of activities of proteases decreased with the increase of Cr content in the substrates. When the Cr contents in elastin or collagen fiber reached 0.5% or 2.0%, the corresponding activity approached to the lowest level, respectively. Different protease showed different activity decrease extend towards tanned protein fibers. Meanwhile, the bating effectiveness of proteases on wet blue was positively related to their activities towards the chrome-tanned protein fibers measured by the established methods, i.e. proteases with highly chrome-tanned elastin and collagen fiber activities could significantly improve the softness of leather. The new methods can be used as an available tool to correctly select proteases and optimize process parameters for wet blue bating. Note de contenu : - MATERIALS AND METHODS : Materials - Methods. Sulfure de sodiumIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Cattle hide was depilated by sodium sulphide and enzyme respectively. The histological structures during the unhairing stages were oberved by transmission electron microscope (TEM). The results showed that for sodium sulphide unhairing, the depilation began with the hydrolyzation of cytoplasm. The epidermis was damaged severely in the first 20 minutes and the epidermis could be dissolved completely in 60 minutes. The Basement Membrane (BM) and desmosome maintained integrity during the whole process. In enzymatic unhairing, the non-keratinised cells were seriously destroyed. The lamina lucida of BM was not noticeable after 30 minutes enzymatic treatment, while the lamina densa did not disappear until 120 minutes.
