In a previous publication from this laboratory, a simple, precise procedure was described for viscometric assay of collagenase activity. While applicable under usual conditions, the procedure fails with samples containing high concentrations of salt. High concentrations of NaCl are present in media in which halophilic microorganisms are grown, and assay of these media for collagenase activity was needed in our study of damage inflicted on salted hides by the halophiles. We here describe a simple modification of the original procedure which permits assay of samples containing as much as 4 M NaCl. The modification, in essence, adds only a single step in which the sample is diluted, prior to the measurement of its viscosity, to reduce its NaCl concentration to £ 0.2 M. The revised procedure provides researchers and leather manufactures with the ability to measure the production of collagenase activity by halophiles. |
