Réaction en chaîne par polyméraseIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Hides and skins are protein based materials containing high amounts of water, which makes them a nutritionally rich media ideal for bacterial growth. Being also the essential raw materials in leather manufacture, it is vital to carefully protect them against bacterial action especially in the early wet processing stages. It is well known that bacterial action can seriously damage the fiber structure of hides and skins. Hair slip, red discoloration and grain pilling are typical examples of bacterial action can lead to damage with subsequent defects on the tanned leather. The present study identifies a new defect caused by bacteria known as "biofilm" on hides and skins. For investigation of the observed defective areas, two techniques were used : Scanning Electron Microscopy (SEM), and Real-Time Polymerase Chain Reaction (RT-PCR). The latter technique, RT-PCR, is a novel rapid method for unique detection, identification and quantification of microorganisms. The results, obtained by applying these techniques, verified the presence of "microbial biofilm" on the leather. Note de contenu : - Material

In soaking various bactericides are used in order to avoid possible defects, these chemicals which are lethal to bacteria may also be harmful to the environment and human health. As an alternative to these chemical based bactericides, the possible use of ozone - which is used as an antimicrobial agent in various industries - in the leather industry is investigated. The effect of ozone applications of 5, 10 and 15 minutes per hour were investigated comparing it to the effect of a sodium dimethyl dithiocarbamide based bactericide. 15 minutes of ozone application has been found to be the most effective on bacterial growth prevention. Cuirs et peaux -- DéfautsIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : This study examined some characteristics of split suede leathers that were treated with a range of Aloe vera concentrations. Following the tanning process of split suede leathers, Aloe vera was applied at five different concentrations (2%, 4%, 6%, 8% and 10%). Leathers treated with Aloe vera took on a yellowish-brown color. Also, the moisture content and softness of the leathers increased according to the concentration of Aloe vera used, and the highest levels of both moisture content and softness were obtained at 8% and 10% application concentrations. The leathers that were treated with 6% or more Aloe vera had slightly higher tear load values when compared with control leathers. In addition, ethanol-based and water-based extracts of Aloe vera showed antimicrobial properties against some Gram (-) and Gram (+) bacteria and C. albicans. Some chemical analyses were made and amino acid composition of collagen hydrolysate obtained from chrome shavings determined. This collagen hydrolysate was used for the production of keratinase from Bacillus subtilis ATCC 6633 as a carbon and nitrogen source. The culture grew in fermentation medium containing various proportion of collagen hydrolysate (1-5% w/v) at pH 7.0 and pH 10.0 with agitation rate of 200 rpm. The best keratinase production was obtained from the medium containing 1% hydrolysate (w/v) at pH 7.0 for 36 hours and with 3% hydrolysate (w/v) at pH 10.0 for 24 hours.
