Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : In order to observe the change of band pattern of collagen fibrils of hides and skins during the leather making process with transmission electron microscope, the effect of fixing and staining on the band pattern was studied first. Collagen fibrils obtained from rat tail tendon and SLS (Segment Long Spacing) - aggregates prepared from acid soluble tail tendon collagen were fixed with formaldehyde, glutaraldehyde, osmium tetroxide or chrome alum, and then positively or negatively stained with phophotungstic acid or uranyl acetate. In general, double positive staining gave far sharper and clearer images than single staining. When positively stained, collagen fibrils usually showed the period consisting of 11 or 12 bands, and contrast and sharpness of each band different according to the fixing method used. The sharpest and clearest images were obtained by fixing the fibrils with chrome alum. There was a great difference between the band pattern of SLS-aggregates fixed with chrome alum and those fixed with glutaraldehyde.

Température de retraitIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : While the chemical composition of a substance is conveniently determined by analytical chemical means and the gross structure assessed by microscopical techniques, the borderline area—ranging from tens of ángströms to a few millimicrons—could well be investigated using an electron microscope. One can not only observe the various entities and study their morphology but also follow the reactions that a substance undergoes when subjected to several treatments.
The advantages of modern electron microscopy include, apart from the require ment of small quantities of sample, stereoscopic view, possibility of heating or cooling the sample, and observation of the specimen under moist conditions.
The results obtained from electron microscopical analyses can broadly be discussed under the following headings :

Ce problème préoccupe, sous un autre aspect, les tanneurs et utilisateurs de peaux de crocodile qui risquent dans un avenir relativement proche de se trouver privés d'une matière première très appréciée. Aussi l'élevage de ces reptiles apparaît-il comme une solution propre à satisfaire à la fois les spécialistes de l'écologie, les industriels du cuir et leurs clients. Il restait à démontrer que cet élevage est possible et à trouver la justification des investissements nécessaires pour le mener à bien. C'est pourquoi le Centre Technique du Cuir a, sur la demande des professionnels, organisé un élevage expérimental de Crocodiles du Nil sur le territoire de la République du Tchad.

Cuirs et peaux -- ConservationIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : Multidrug-resistant Enterobacteriaceae originating from animals' intestinal tracts may be found on salted and soaked hides/skins. The presence of proteolytic, lipolytic, multidrug-resistant Enterobacteriaceae on hides/skins and the resulting destructive processes results in economic lusses to the leather industry and creates health hazards for workers. To minimise this destructive bacterial presence, the bactericidal effect of combined application of direct or alternating electric current and an antibacterial agent against multidrug-resistant Enterobacteriaceae were examined in this study.
Multidrug-resistant Citrobacter freundii, Citrobacter koseri, Enterobacter cloacae, Serratia rubidaea, Serratia marcescens, Serratia plymuthica, Morganella morganii, Proteus mirabilis, Providencia rettgeri, isolated from soaked hides/skins, were used in the present study. Among the test isolates, Proteus mirabilis, Serratia marcescens, Serratia plymuthica, Serratia rubidaeai were protease and lipase producer strains. Ail test isolates were resistant to critically important antimicrobials which are used in both human and veterinary medicine. Bacterial effect of combined application of 508mA/cm2 direct electric current, 454mA/cm2 alternating electric current and an antibacterial agent (2- (thiocyanomethylthio) benzothiazole) against the mixed culture of these micro-organisms were investigated in nutrient broth containing 3% NaCI. After application of six cycles of the combine electric current treatment against the mixed culture of multidrug-resistant Enterobacteriaceae treated with (2-(thiocyanomethylthio) benzothiazole) was completely killed within five hours. The Log10 reduction of the mixed culture at the end of the experiment was T55.
In conclusion, the combined application of direct electric current, alternating electric current and antibacterial agent may be used in soak liquors to eliminate proteolytic, lipolytic, multidrug-resistant Enterobacteriaceae in the leather industry.Note de contenu : - Test micro-organisms

Wet-blue (tannage)Peau tannée au chrome (le chrome donne une couleur bleue)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Inductively Coupled Plasma Optical Emission Spectrophotometer (ICP-OES) was used to accurately determine the Chromic Oxide content in 90 total Wet Blue, Crust, and Finished Leather samples. Accuracy was verified on each sample by testing retained composite material using ASTM D2807 Perchloric Digestion / Sodium Thiosulfate Titration Method. The ICP-OES Method correlated to ASTM D2807 with less than 2% deviation from the averaged mean value and less than 3% deviation from the averaged standard deviation value. The Chromic Oxide mean values for ASTM D2807 and ICP-OES were 3.11 ± 0.70% and 3.17 ± 0.68% respectively. The new method maintains a highly consistent statistical capability with Cpk at 2.15 vs. 2.17. Oxidation of chromium in leather samples is not required for chromic oxide detection. Perchloric acid more than doubles the required consumables cost, test time and hazard required to validate chromic oxide levels in leather materials. The ICP-OES method eliminates the crystal formation explosion hazard and chlorinated waste environmental hazards associated with Perchloric acid. Replacing ASTM D2807 with the proposed method by tannery validation laboratories is cheaper, faster, safer and more environmentally sustainable than titration by our classic technique.
