Réticulation (polymérisation)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Genipin(GP) has been used as a sort of perfect naturel cross-linking agent for collagenous biomaterials, but the cross-linking mechanism between collagen and GP is not clear. The aldehyde groups can be formed in aqueous solution of Genipin (GP), which can react with Cu(OH)2 and be used in the silver mirror reaction. About 0.2506 moi aldehyde groups were determined by Decolor Fuchsin Spectrophotometry in 1mol GP. In neutral solution, the ultraviolet absorption peak of GP appeared at 242.10nm, whereas in alkaline solution, it GP moved to 271.70nm due to the generation of enolates structure. Compared with the neutral condition, the GP molecular size almost doubled in alkaline solution, therefore il was speculated that dimers were formed, In this work, porcine acellular dermal matrix (pADM) was employed as a model of collagen to react with Genipin. The colour of GP cross-linked pADM (GP-pADM) turned into yellow from white initialty and finally into dark blue and a new absorption peak was round at 2356.79cm-1. With an increasing amount of GP, the shrinkage temperature (Ts) of GP-pADM increased slowly from 55.4°C to 80°C (6% dosage). Regarding the formation of dimers, the size of GP molecule was increased which fitted the distance of two aminos of the collagen for cross-linking. Furthermore, due to the ring structure of genipin and cross-linking bonds, the amount of rigid structures of collagen increased, consequently improved collagen stability was achieved. Fibroblasts grew well on GP-pADM with great morphology, and the vitro cytotoxicity was grade 0. Stabilité thermiqueIndex. décimale : 675 Technologie du cuir et de la fourrure Résumé : A novel method for the immobilization of an enzyme was established by using solid waste from tanneries as the carrier. A series of inorganic and organic carriers with good behaviours were evaluated in this context. The results showed that the enzyme immobilization efficiency was optimal when the enzyme offer, reaction temperature, pH, ionic strength and time were set at 335mg enzyme per g carrier, 25°C, 6.5, 0.1mol/L and 60 minutes, respectively. The optimal temperature and pH of the immobilized enzyme in terms of protease were 55°C and 8.0, respectively, similar to that for the free enzyme. The immobilized enzyme remained 64% of its original activity after keeping at 55°C for 3 hours, while the free enzyme lost nearly 54% under the same conditions, indicating the improvement of enzyme thermal stability after enzyme immobilization. Furthermore, the leather industry oriented application of the immobilized enzyme was intended for the dehairing of cattle hide. Note de contenu : - EXPERIMENTAL PROCEDURES : Materials - Methods

Immunisation (épilage)Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : To better understand the mechanism of alkali-induced hair immunization, in this work native bovine hair was selected as the direct research subject rather than fresh salted bovine hide. The state of hair was evaluated, both before and after having undergone the procedure of immunization by measuring the changes of active groups, thermal stability, surface morphology, strength, calcium content and cystine content. The result indicates that the immunization is indeed caused by the interaction between calcium hydroxide and the cystine active center. Meanwhile, the optimum condition for immunization was: calcium hydroxide 15g/I and treatment time 90 minutes.
