Collagen is the major protein present in the skins and hides in association with various globular proteins, viz. albumin, globulin, mucoids; and fibrous proteins such as elastin, keratin, and reticulin. The hide or skin, once flayed, has cells that are deprived of supply of oxygen and nutrients. Hides and skin are getting damaged mainly by bacteria, which are responsible for the decomposition of untanned proteins (in raw hides and during soaking), while fungi thrive on leathers containing carbohydrates, fats and proteins. We have characterized the bacteria present in skin samples and found that the bacterial strains with proteolytic and lipolytic activities were predominant. Based on the biochemical-physiological characteristics, they were identified as Gram-positive rods and cocci such as Bacillus subtilis, B. megaterium, and B. cereus and Staphylocorcus epidermidis, and S. aureus and Lactobacillus acidophilus and L. fermentum. In addition, nine strains of Micrococcus luteus and one strain as Streptococcus feacalis were also identified. Total of sixteen Gram-negative bacterial strains, 3 were identified as Neisseria, 7 as Proteus sp., and 6 as Pseudomonas sp. Qualitative assessment of the degradation of collagen fibers leading to skin damage suggested that Gram-positive organisms with proteolytic activity are mainly involved in the putrefaction of goat/ sheep skin.
