The enzyme was found to be thermostable and alkali stable. This enzyme was used for the hydrolysis of chrome shavings. Pretreatment study was done on chrome shavings prior to enzymatic hydrolysis. An efficient removal of chrome and protein hydrolysate from chrome shavings was observed. The currently employed tanning methods lead to considerable waste of chromium(III) salts used. There is a need to develop mixed-metal complexes of chromium to reduce the use of chromium in tanning and avoid recovery-reuse methodologies required in conventional chrome tanning. A mixed-metal complex of chromium and zirconium has been developed. The mole ratios of zirconium and chromium in the complex have been optimized to achieve above 100 °C shrinkage temperature, without loss. of leather quality and changes to conventional tanning methods. Therefore, hexavalent chromium is not likely to be present in leather. Nevertheless, Cr(VI) has been reported to be present. At the moment it is not clear whether the positive test results are indeed the consequence of hexavalent chromium extraction from leather or not. Despite this, Cr(VI) formation during extraction has been suggested as a probable reason for most of the positive test results. In the present investigation, leather analyses were carried out using different buffer solutions and also the same buffer solution at different pH values. Analysis for Cr(VI) was carried out under simulated extraction conditions also. The results of all these experiments indicate that the amount of Cr(VI) formed during extraction is too little to be considered as an important reason behind testing that yields positive findings of Cr(VI). Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Prior research from this laboratory has demonstrated that commercial gelatins can be modified with a microbial transglutaminase with a resulting effect on their physical properties. The gels isolated from those treatmentshad improved physical properties and films prepared from these modified products had superior mechanical properties. In this present study we modified commercial gelatins and experimental gelatin products isolated from chromium-containing tannery waste and examined the functional properties as well as their molecular weight distributions. On increasing the enzyme concentration, the gelatins become highly polymerized, some to such high molecular weight that the moieties will either not go into the SDS-PAGE gel or, because of their insolubility, cannot even be applied to the gel. Physical, foaming, adhesive and water absorption properties of enzymatically-modified gelatins were improved. The extent of modification of the resulting products was demonstrated by the molecular weight distribution studies and these correlated with changes in physical and functional properties. A particular advantage to using this enzymatic modification was the improvement in physical properties imparted to low quality gelatins. The modified protein has applications in a wide range of products, including adhesives, cosmetics, films, encapsulating agents, fertilizers, and--of significance to the leather industry--as coatings or fillers.
