Revêtements organiquesIndex. décimale : 667.9 Revêtements et enduits Résumé : Our study decided to use the new and revolutionary approach in the field of pharmaceutical coating processes called the artificial neural network (ANN) by using the neural networks toolbox derived from the Matlab® software. The experiments were performed using tablets of Alfuzosin Chlorhydrate as a model filler, and an aqueous solution of Surelease as a polymer in different contents. The various parameters that can affect coating thickness, weight gain, and the coefficient of variation CV, such as spray rate, air pressure, solid content, speed of the drum, pan loading, and time of coating, were studied. The properties of the coated tablets were evaluated using the ANN, and both the parameters of the coating process and the properties of the coated tablets were used as a basis for optimization, as well as the choice of the optimal structure of the ANN model. It was found that the best neural network architecture had 7 neurons in the hidden layer, with a mean square error of 3.515 and a determination coefficient of nearly 1. The relative importance of each independent variable was quantified using the Garson equation. In this study, spray rate was found to have the highest impact on the properties of tablets. Revêtements poudreIndex. décimale : 667.9 Revêtements et enduits Résumé : Film coating of tablets is the application of a thin layer of coating material to improve the tablet properties. Different coating materials like hydroxypropyl methylcellulose (HPMC) and polyvinyl alcohol (PVA) have been widely used in tablet coatings for immediate release of the active pharmaceutical ingredient. A new immediate release film coating formula that contains modified corn starch as the coating polymer was investigated, and it shows improved physiochemical properties over conventional film coating material (HPMC and PVA). The purpose of this study is to investigate the potential of the new modified starch coating as a coating material in comparison with HPMC- and PVA-based formulas. Evaluation was done on a coating powder dispersion of 20% w/w by measuring the viscosity, sedimentation, foaming behavior, and microbial stability. Free films were prepared from each coating formula and tested for water vapor permeability and morphology. The coating process was then executed on the model tablets (levofloxacin 500 mg) after optimizing the coating parameters. Characterization of modified starch film-coated tablets was done by testing morphology, hardness, friability, thickness, disintegration, and drug release profiles. It was concluded that the new modified starch coating is superior to HPMC and PVA formulations in terms of free film and applied coated tablet esthetics, inner structure composition, preparation and coating process parameters. Tablet coating is carried out for the purposes such as decoration and protection of tablets along with some functional properties. The coating is a homogeneous mixture of polymeric material ; a very thin layer is applied on a substrate of the drug by using various techniques. These coatings techniques are either manually operated or fully automated. The automatic process gives coating with more uniform film properties at a high production rate. The coating is the last unit operation in the manufacturing of drugs in the pharmaceutical industries. Tablet coating involves the use of various binders, colour forming agents (colorants), organic solvents and plasticizers to get the required characteristics. Coating a tablet is always a great challenge to coating a formulator as it involves various statutory requirements to qualify as well as technically it also performs well against various environmental changes. Coating plays a key role in the pharma industry to protect the drugs from different environmental challenges like changes in humidity, light, heat etc. Now a days in coating application industries various new coating techniques are available like spray and fluidised bed methods that are developed to overcome the limitations of old techniques. SorbitolIndex. décimale : 667.9 Revêtements et enduits Résumé : The aim of the present study was to develop pegylated cellulose acetate coating layer having leachable membrane created by controlled porosity osmotic pump (CPOP) technique. Already optimized core tablets of Captopril were used for the application of CPOP. In Box–Behnken design, cellulose acetate, polyethylene glycol, and sorbitol were used as variables and %Captopril release as a response for the coating process. Formulations were characterized by weight gain, thickness of leachable membrane, content uniformity, drug release, pH, dissolution medium osmolality effect, FTIR, and stability analysis for the optimization purpose. Optimized formulation was within the pharmacopoeial limits. Observed weight gain was 4.38–6.97%, and thickness of leachable membrane was 1.24–1.47 mm. Content uniformity was found to be 91.65–99.61%. Phosphate buffer of pH 6.8 showed 86.22–92.79% drug release and the first-order release pattern. pH-independent but osmolality-dependent drug release was observed. No incompatibility between the ingredients of prepared dosage form was observed in FTIR analysis. Accelerated stability study for 6 months showed no significant change in the prepared dosage form. Conclusively, prepared CPOP tablets can be used for the controlled release of Captopril in hypertensive patients to maintain the desired drug concentration within the body for required time period.
