Tannage Résumé : The effect of incremental increases in concentration of sulfates on anaerobic digestion was investigated using a laboratory-scale Upflow Anaerobic Sludge Blanket (UASB) reeactor fed semi-continuously with a synthetic wastewater. A concentration of up to 6000mg/l SO42- was found to have no effect on an acclimatized anaerobic digester with the exception of a decrease in the methane yield. The sulfides formed as a result of microbial sulfate reduction were distributed between soluble sulfides and hydrogen sulfide in the biogas. Precipitation of heavy-metal sulfides also occured in the digester. These results suggest that the concentration of sulfates and sulfides present in tannery wastewaters would be insufficient to cause failure of anaerobic digesters. Tannage Résumé : Research involving selected combinations of commercial proteases provided evidence that a synergistic depilatory effect occurs when proteases of complementary bond specificities are used in conjunction in enzymic depilatory preparations. Some form of strong alkali treatment of skins was found to be necessary to produce a skin fibre matrix of the prerequisite quality when the skin was depilated by proteases since, otherwise, the depilated skin after acetone dehydration was found to be depleted and stiff. Calcium hydroxide alone was found to be inadequate for this task, probably owning to the fact that it is less alkaline than the lime-sulfide mixture. Sodium hydroxide was therefore used in conjunction with calcium hydroxide (lime) to match the alkalinity of the lime-sulfide solution in order to produce skin characteristics comparable with those from a line-sulfide unhairing treatment.
A combinaison of the information provided by the results obtained in the present systematic study combined with information gleaned from the relevant literature allowed for the construction of a model to represent the possible mechanism of enzymic depilation of skins. The Proteus vulgaris bacterium apparently produced a single protease or protease complex according to ion exchange and gel exclusion chromatography. The protease had maximal proteolytic activity in the 8.0-9.0 pH unit rage and was quite stable at high salinities (1.0 M). The protease was heat labile being increasingly inactivated when exposed to temperatures above 30°C ; the inclusion of divalent calcium ions failed to significantly thermostabilise the protease. The protease had a molecular mass of 44 000 daltons. Tannage Résumé : The ability of anaerobic microbial populations to adapt to high sodium chloride concentrations was investigated. It was found that a sodium chloride (NaCl) concentration of up to 17.000 mg/L had no inhibitory effects on the anaerobic digestion process. There were no notable differences in digester performance relative to a control with the exception of an increase in the methane content of the biogas. These results suggest, therefore, that the concentration of NaCl present in tannery wastewaters, which ranges from 2.500 to 12.500 mg/L, would have no significant effect on an acclimatized anaerobic digester treating these wastes. It was found that an influent concentration of 5000mg/l trivalent chromium could be tolerated by acclimatized methanogenic bacteria. However,if the concentration in the synthetic waste was further increased, inhibition of the methanogens resulted, with a corresponding decrease in degester performance. These results suggest,therefore, that the concentration of trivalent chromium present in tannery effluents and sludges,which ranges from 1 to 2500 mg/l would have no significant effect on an acclimatized, stable anaerobic digester treating these wastes.
