Langues : Américain (ame) Index. décimale : 675 Technologie du cuir et de la fourrure Résumé : Solid-substrate fermentation(SSF) of an Aspergillus flavus isolate has been developed for the production of an alkaline protease.Since SSF is known to have distinct economic advantages over conventional methods, it is hoped that this alkaline protease will be available at a commercially attractive price.A study of the properties of the enzyme indicate that it is active over a pH range of 6.5 to 10.5, with a temperature optimum at 45°C.It has high caseinolytic activity and no activity against collagen.Evaluation of this enzyme as a depilating agent for goat skins indicate that its dehairing capacity is satisfactory.Physical testing of the finished leathers of both experimental and control groups, indicates that the enzymically dehaired skins had better strength properties when compared to the controls.Miscroscopic evaluation of the pelts showed that enzymic dehairing compared favorably with the lime-and-sulfide method. It has been established that enzymatic unhairing is an environmentally friendly alternative to the conventional process. The enzyme, an alkaline protease (clarizyme) is produced by aspergillus flavus isolate using solid substrate fermentation (SSF). Since SSF has distinct economic advantages over conventional methods, it is hoped that this alkaline protease will be available at a commercially attractive price. Evolution of this enzyme as a depilating agent for goat leather establishes that the enzymatically dehaired skins have better strength compared to the controls. This paper highlights the process development and scaling-up strategies adopted in the enzyme production and its application.
